多西紫杉醇协同卡铂对骨肉瘤细胞MG-63细胞毒性的实验研究

摘要: 目的:研究多西紫杉醇和卡铂联用对骨肉瘤细胞MG-63的增殖抑制作用,寻找新的骨肉瘤临床治疗方案。方法:采用MTT法细胞毒性试验,测定多西紫杉醇及卡铂对MG-63细胞株的抑制率并计算其IC50值,根据所测得的IC50值,分别用IC50多西紫杉醇、IC50卡铂、1/2 IC50多西紫杉醇 + 1/2 IC50卡铂作用于MG-63细胞株24 h、48 h、72 h,比较三组药物处理方法对MG-63细胞株的抑制率。结果:根据相应实验结果制作生长曲线通过公式IC50 = lg − 1[Xm − i(ΣP − 0.5)]求得两药的IC50值分别为0.25 mg/ml和4.00 mg/ml。分别用0.25 mg/ml多西紫杉醇、4 mg/ml卡铂、0.125 mg/ml多西紫杉醇+2 mg/ml卡铂与骨肉瘤细胞株MG-63作用24 h时,其细胞毒性指数(CI)分别为:38.6%、48.7%和60.0%,3组比较差异有统计学意义(P < 0.05);作用48 h时,其细胞毒性指数(CI)分别为:44.3%、49.2%、73.0%,3组比较差异有统计学意义(P < 0.05);作用72 h时,其细胞毒性指数(CI)分别为59.3%、58.6%、79.9%,多西紫杉醇组于卡铂组比较差异无统计学意义(P > 0.05),联用组与另外两组比较差异均由统计学意义(P < 0.05)。结论:半量多西紫杉醇、卡铂联合应用比单独应用多西紫杉醇或卡铂对骨肉瘤细胞株的细胞毒性作用更强。
Abstract: Objective: To research the cytotoxicity of docetaxel combined with carboplatin on osteosarcoma cell line MG-63. Methods: Using MTT assay, we obtained the mean IC50 of docetaxel and caboplatin for the osteosarcoma cell line MG-63. The MG-63 was incubated with IC50 docetaxel, IC50 carboplatin and 1/2 IC50 docetaxel + 1/2 IC50 carboplatin for 24 h, 48 h, 72 h. The cytotoxicity of each group was measured by MTT assays. Result: By the formula IC50 = lg − 1[Xm − i(ΣP − 0.5)] to obtain the IC50 of docetaxel and carboplatin for osteosarcoma cell MG-63 were 0.25 mg/ml and 4.00 mg/ml respectively. The mean cytotoxicity index (CI) of 0.25 mg/ml docetaxel, 2 mg/ml carboplatin and 0.125 mg/ml docetaxel + 2 mg/ml carboplatin for 24 h were 38.6%, 48.7% and 60.0% respectively (P < 0.05); and those for 48 h were 44.3%, 49.2% and 73.0% respectively (P < 0.05); and those for 72 h were 59.3%, 58.6% and 79.9% respectively (the 0.25 mg/ml docetaxel group and 2 mg/ml carboplatin group P > 0.05; the combined group compared with the other two groups P < 0.05). The cytotoxicity index of combined group was significant increased. Conclusion: The combination of half dose docetaxel with carboplatin appears more active as docetaxel or carboplatin alone in full doses in treatment of osteosarcoma cell line MG-63 in vitro.
