    摘要: 目的:了解人体肠道益生菌细菌素的抑菌活性。方法:从健康人粪便中分离肠道益生菌,用传统方法和PCR法鉴定菌株;Yang法提取细菌素,96微孔板法筛选产广谱、高效抑菌作用细菌素菌株;96孔板显色法测定细菌素对金黄色葡萄球菌(S. aureus),肺炎链球菌(S. pneumoniae),大肠埃希菌(E. coli),肺炎克雷伯菌(K. pneumoniae),普通变形杆菌(P. vulgaris),伤寒沙门菌(S. typhi),福氏志贺菌(S. flexneri),铜绿假单胞菌(P. aeruginosa),鲍曼不动杆菌(A. baumannii),白色念珠菌(C. albicans)抑菌效果。结果:分离出32株肠道益生菌,包括4株双歧杆菌(B)、6株乳杆菌(L)和22株肠球菌(E);B3、B4、L5、E12、E15、E18为产广谱高效细菌素菌株;B3、L5、E15菌株所产细菌素对革兰阳性菌和阴性菌均有抑制作用;其中B3、L5细菌素对伤寒沙门菌和肺炎链球菌具有较强的抑制作用;E15细菌素对肺炎链球菌和白色念珠菌有较强的抑制作用。结论:人体肠道益生菌产生的细菌素抑菌谱较广且活性较高,具有一定的开发潜力。Object: To reveal the bacteriostatic activity of bacteriocins produced by human intestinal probiotics. Methods: Intestinal probiotics were separated from excrement of healthy people and identified by routine method and PCR. Bacteriocins were refined as Yang’s way and strains that could produce broad-spectrum efficient bacteriocins were selected by using 96-well plate method. Bacteriostatic activity of every selected bacteriocin was detected on S. aureus, S. pneumoniae, E. coli, K. pneumonia, P. vulgaris, S. typhi, S. flexneri, P. aeruginosa, A. baumanni and C. albicans by 96-well plate method respectively. Results: 32 strains of intestinal probiotics were separated, including 4 bifidobacteria (B), 6 lactobacillus (L), and 22 enterococcus (E). B3, B4, L5, E12, E15, E18 were broad-spectrum efficient bacteriocin producing strains. Bacteriocin of B3, L5, E15 has bacteriostatic action on gram positive and negative bacteria. Both B3 and L5 bacteriocin has strong inhibitory action on S. typhi and S. pneumoniae, while E15’ shows higher inhibitory effects on C. albicans and S. pneumoniae than other bacteria used in this study. Conclusion: Bacteriocins produced by intestinal probiotic bacteria have broad bacteriostatic activity, and some of them are high efficient.
