口腔潜在恶性病变唾液生物标记物对口腔鳞癌的癌前预测
Precancerous Prediction of Oral Squamous Cell Carcinoma by Salivary Biomarkers in Potential Oral Malignant Lesions
摘要: 目的:本研究通过比较口腔鳞状细胞癌(OSCC)、口腔潜在恶性病变(OPMD)及对照组的唾液中IL-6、IL-8、MMP-9的浓度差别,筛选出能够用于预测OSCC发生的特异性的唾液分子标记物。方法:将60名受试者分为3个研究组,即OSCC组(n = 20)、OPMD组(n = 20)和健康对照组(n = 20)。所有受试者均提供非刺激性的唾液样本,使用ELISA试剂盒检测IL-6、IL-8、MMP-9的浓度。结果:OSCC组的唾液IL-6、IL-8、MMP-9浓度显著高于对照组(P < 0.05),OPMD组的唾液IL-6、IL-8、MMP-9浓度高于对照组,且IL-8的浓度具有统计学差异(P < 0.05)。OSCC组的唾液IL-6、IL-8、MMP-9的浓度显著高于OPMD组(P < 0.05)。结论:本研究表明OSCC唾液中的IL-6、IL-8、MMP-9的浓度相比对照组显著增高,且OSCC和OPMD组间唾液IL-6、IL-8、MMP-9的浓度存在统计学差异。唾液IL-6、IL-8、MMP-9在OSCC和OPMD的治疗、癌前预测及随访中是一种有临床参考价值的特异唾液分子标志物。
Abstract: Objective: To screen specific salivary biomarkers that can be used to predict the occurrence of OSCC by comparing the concentrations of IL-6, IL-8 and MMP-9 in saliva of oral squamous cell carcinoma (OSCC), oral potential malignant disease (OPMD) and control group. Methods: 60 subjects were di-vided into three study groups, named OSCC group (n = 20), OPMD group (n = 20) and healthy con-trol group (n = 20). All subjects provided non irritant saliva samples, and detected the concentra-tions of IL-6, IL-8 and MMP-9 with ELISA kit. Results: The salivary concentrations of IL-6, IL-8 and MMP-9 in OSCC group were significantly higher than those in control group (P < 0.05). The salivary concentrations of IL-6, IL-8 and MMP-9 in OPMD group were higher than those in control group, and the concentration of IL-8 was statistically different (P < 0.05). The saliva concentration of IL-6, IL-8 and MMP-9 in OSCC group was significantly higher than that of OPMD group (P < 0.05). Conclusions: This study shows that the saliva concentration of IL-6, IL-8 and MMP-9 in OSCC is significantly higher than that of the control group, and the saliva concentration of IL-6, IL-8 and MMP-9 between OSCC and OPMD groups is statistically different. Salivary IL-6, IL-8 and MMP-9 are specific salivary bi-omarkers with clinical reference value in the treatment, precancerous prediction and follow-up of OSCC and OPMD.
