益气类中药对博来霉素致肺纤维化大鼠模型气道重塑的影响
Effects of Qi Invigorating Herbs on the Airway Remodeling of Bleomycin Induced Pulmonary Fibrosis in Rats Model

摘要: 目的:观察博莱霉素(BLM)所致肺纤维化模型大鼠肺组织病理形态和肺组织中转化生长因子(TGF-β1)的表达水平,探讨益气类中药对肺纤维化的作用及其机制。方法:32只健康雄性SD大鼠随机分为空白组、模型组、益气组、泼尼松组,采用气管内滴加博莱霉素的方法制作肺纤维化模型,并应用相应的药物灌胃干预,各组大鼠分别于第28 d处死,肺组织切片经HE染色后观察病理形态,ELISA法测定大鼠肺组织中TGF-β1的表达水平。结果:与空白组比较,其余各组大鼠肺组织中TGF-β1表达明显增强(P < 0.01);两治疗组肺纤维化程度较模型组明显减轻(P < 0.01)TGF-β1表达也明显低于模型组(P < 0.01);益气组肺纤维化程度较泼尼松组轻(P < 0.05)TGF-β1表达也明显低于泼尼松组(P < 0.01)结论:益气类中药能够抑制TGF-β1表达,干预肺纤维化大鼠的气道重塑,抑制或延缓肺纤维化的发生和发展,疗效较泼尼松更好。
Abstract: Objective: To observe the tissue pathology and expression level of transforming growth factor (TGF-β1) of the bleomycin (BLM) induced pulmonary fibrosis rats model lung, to explore the effect of Qi invigorating herb on pulmonary fibrosis and its mechanism. Methods: 32 healthy male SD rats were randomly divided into blank group, model group, Yiqi group and prednisone group, making the pulmonary fibrosis model by intratracheal bleomycin plus, and applying the corresponding drug stomach intervention, the rats were sacrificed at 28 d, observing the lung tissue slice pathological morphology after HE staining, and determining the expression level of TGF-β1 in lung tissue of rats with ELISA method. Results: Compared with the blank group, other groups in the lung tissue of rats with TGF-β1 expression were obviously increased (P < 0.01); in the two treatment groups, pulmonary fibrosis was significantly reduced compared with model group (P < 0.01), the expression level of TGF-β1 was significantly lower than that in the model group (P < 0.01); Compared with prednisone group, the degree of pulmonary fibrosis rats was lighter (P < 0.05) in Yiqi group, the expression level of TGF-β1 was significantly lower than the prednisone group (P < 0.01) too. Conclusion: Qi invigorating herbs can inhibit the expression of TGF-β1, intervene the airway remodeling in rats with pulmonary fibrosis, inhibit or delay the occurrence and development of pulmonary fibrosis, and have a better effect than prednisone.
