Abstract: AIM: To investigate the effect of As2O3 on the expression of telomerase reverse transcriptase mRNA in ACC-2 cells. METHODS: ACC-2 cells were cultured. The different drug concentration gradient (0, 1.0, 2.0, 4.0, 8.0 μmol/L) of As2O3 were applied to ACC-2 cells for 24 h and 48 h respectively. The hTERT mRNA expression of ACC-2 cells before and after As2O3’s induction was detected by RT-PCR. RESULTS: RT-PCR results showed that hTERT mRNA expression was significantly higher in the control group, while after As2O3 action to ACC-2 cell for 24 h and 48 h, with the increase of As2O3 concentration (1.0 μmol/L, 2.0 μmol/L, 4.0 μmol/L, 8.0 μmol/L), hTERT mRNA expression decreased gradually (P < 0.01). CONCLUSION: As2O3 can induce apoptosis of ACC-2 cells by downregulating hTERT mRNA transcript expression.
