摘要: 人类乳头瘤病毒(human papillomavirus, HPV)是小型双链环状DNA病毒,是世界上最常见的性传播病毒。高危型HPV (high-risk HPVs, HR-HPV)持续感染可导致宫颈上皮内瘤变(Cervical Intraepithelial Neoplasia, CIN)与宫颈癌的发生。除了众所周知的转化特性外,HPV感染的细胞还会主动指导局部环境,并创造一个支持性的感染后微环境(post-infection microenvironment, PIM),这被认为是病毒持续、传播和恶性进展的关键因素。从HPV感染到发生癌前病变、直至发展为宫颈癌是一个长期持续的过程,了解PIM有助于探讨HPV持续感染并导致CIN及宫颈癌的可能机制,可为宫颈癌的防治提供新思路。本文总结了目前对PIM中关键信号通路、成分的理解,就HPV感染后微环境的变化进行综述。
Abstract: Human papillomavirus (HPV) is a small double-stranded circular DNA virus that is the most com-mon sexually transmitted virus in the world. Persistent infection of high-risk HPVs (HR-HPV) can lead to Cervical Intraepithelial Neoplasia (CIN) and cervical cancer. In addition to its well-known transforming properties, HPV-infected cells actively direct the local environment and create a supportive post-infection microenvironment (PIM), which is believed to be a key factor in the con-tinued spread and malignant progression of the virus. It is a long and continuous process from HPV infection to precancerous lesions and development to cervical cancer. Understanding PIM is helpful to explore the possible mechanism of HPV persistent infection leading to CIN and cervical cancer, and can provide new ideas for the prevention and treatment of cervical cancer. This paper summarizes the current understanding of key signaling pathways and components in PIM, and re-views the changes of the microenvironment after HPV infection.
