摘要: 目的:细胞焦亡是阿尔兹海默症相关的重要炎症过程。本研究在已有研究的基础上,进一步筛选鉴定阿尔兹海默症中细胞焦亡的潜在生物学靶点。材料方法:首先,从基因表达综合数据库中筛选mRNA表达谱数据集GSE5281,并使用R软件(4.0.0版)分析阿尔兹海默症和细胞焦亡相关的潜在差异表达基因。随后,对选定的候选基因进行蛋白质–蛋白质相互作用分析、基因本体富集分析和京都基因与基因组百科全书通路富集分析等。并最终根据hub基因的排名,筛选出可能的潜在靶向基因。结果:差异性分析确定了768个可能的与阿尔兹海默症相关的潜在靶点。通过开放的细胞焦亡数据库,我们进一步筛选出76个细胞焦亡相关的差异基因。蛋白质–蛋白质相互作用网络展示出绝大多数候选靶点存在密切的相互作用。并筛选出了网络中核心的hub基因。结论:我们的研究鉴定了76个阿尔兹海默症中潜在的细胞焦亡相关的差异表达基因。并借助蛋白质互作网络总结出可能最值得关注的前10个hub基因(SOD1、YWHAB、TUBB4B、TUBB、TUBA1C、GJA1、GAPDH、HSP90AB1、HSPA8和YWHAZ)。本研究结果可能有助于进一步了解阿尔兹海默症的发病机制,并指导治疗。
Abstract: Objective: Pyroptosis is an important inflammatory process associated with Alzheimer’s disease (AD). On the basis of previous studies, this study aims to further screen and identify the potential biological targets of pyroptosis in AD. Material Method: Firstly, the mRNA expression profile dataset GSE5281 was screened from the Gene Expression Omnibus database, and the potential differentially expressed genes related to Alzheimer’s disease and pyroptosis were analyzed by R software (version 4.0.0). Subsequently, the selected candidate genes were subjected to protein-protein interaction analysis, Gene Ontology enrichment analysis, and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis. Finally, according to the ranking of hub genes, the possible potential target genes were screened out. Result: Differential analysis identified 768 potential AD targets. Through the open pyroptosis database, we further screened 76 differentially expressed genes related to pyroptosis. The protein-protein interaction network showed that the vast majority of candidate targets had close interactions. The core hub genes in the network were screened out. Conclusion: Our study identified 76 differentially expressed genes potentially involved in pyroptosis in AD. The top 10 hub genes (SOD1, YWHAB, TUBB4B, TUBB, TUBA1C, GJA1, GAPDH, HSP90AB1, HSPA8 and YWHAZ) were summarized by protein-protein interaction network. The results of this study may contribute to further understanding of the pathogenesis of AD and guide treatment.
