Abstract: Gastric cancer remains a malignancy with significant global morbidity and mortality. Peritoneal metastasis is a common mode of dissemination and a primary contributor to poor patient prognosis. The S100 family of proteins, a group of calcium-binding proteins, plays a crucial role in tumor initiation, progression, and metastasis. This review comprehensively focuses on the fundamental theories, epidemiology, diagnostic techniques, therapeutic strategies, controversies, and future perspectives regarding S100 proteins in the context of gastric cancer peritoneal metastasis. Through a synthesis of relevant literature, it elucidates the structural and functional characteristics, molecular mechanisms, epidemiological features, diagnostic value, therapeutic potential, and existing controversies associated with S100 proteins in this process. Furthermore, it offers insights into future research directions, aiming to lay a theoretical foundation and provide novel perspectives for the precise diagnosis and treatment of peritoneal metastasis in gastric cancer.
