Abstract: O-linked β-N-acetylglucosamine (O-GlcNAc) glycosylation is a dynamically reversible protein post-translational modification that is present on serine or threonine residues of thousands of proteins in cells and plays an important role in various cellular processes by regulating transcription, epigenetics, and signaling. Diabetes is a chronic metabolic disease, and long-term hyperglycemia can affect a variety of tissues and organs, leading to a series of complications. In recent years, accumulating evidence has indicated that O-GlcNAc glycosylation is extensively involved in the occurrence and development of diabetes and its complications. This article reviews the regulatory mechanisms and physiological functions of O-GlcNAc glycosylation, with a focus on its mechanistic roles in diabetes and its complications.
