摘要: 肝细胞癌(hepatocellular carcinoma, HCC)是常见的恶性肿瘤之一,其发病机制复杂,临床治疗仍面临诸多挑战。近年来,AKT信号通路及线粒体功能异常在HCC发生发展中的作用逐渐受到关注,其中线粒体通透性转换(mitochondrial permeability transition, MPT)被认为是调控细胞命运的重要环节。与此同时,来源于天然产物的姜黄素因其多靶点生物学效应,在肿瘤干预中的潜在价值不断被报道。本文围绕AKT-MPT轴在HCC中的作用机制进行综述,重点从AKT信号调控、线粒体通透性转换的分子基础以及CypD相关调控机制等方面,对相关研究进展进行了梳理。在此基础上,进一步总结了姜黄素在调控AKT信号及诱导线粒体通路介导细胞凋亡中的可能作用,并对其潜在的抗肿瘤机制进行了分析。总体来看,AKT-MPT轴可能在HCC细胞存活与凋亡调控中发挥重要作用,而姜黄素通过多途径调控该信号网络,因此展现出一定的应用前景。但目前相关机制研究仍存在一定限制,其临床转化价值仍有待进一步探讨。
Abstract: Hepatocellular carcinoma (HCC) is one of the most common malignant tumors, with complex pathogenesis and limited therapeutic options. In recent years, increasing attention has been paid to the role of AKT signaling and mitochondrial dysfunction in HCC progression. Among these, mitochondrial permeability transition (MPT) has been considered an important regulatory event in determining cell fate. Meanwhile, curcumin, a natural polyphenolic compound, has attracted interest due to its multi-target biological activities and potential anti-tumor effects. This review summarizes the current understanding of the AKT-MPT axis in HCC, with a focus on recent research advances in AKT signaling regulation, the molecular basis of mitochondrial permeability transition, and cyclophilin D (CypD)-related regulatory mechanisms. In addition, the potential effects of curcumin on modulating AKT signaling and inducing mitochondrial-mediated apoptosis are discussed, and its potential anti-tumor mechanisms are analyzed. Overall, the AKT-MPT axis may play a critical role in regulating cell survival and apoptosis in HCC. Curcumin appears to interfere with this signaling network through multiple mechanisms, showing promise for clinical applications. However, current research on the underlying mechanisms remains limited, and its clinical translational value requires further investigation.
