摘要: 目的:探讨天麻素对1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)诱导的PD小鼠的运动功能的保护作用。方法:选用SPF级雄性C57BL/6小鼠,随机分为MPTP PD模型组及MPTP + 天麻素组,每组5只。采用MPTP 25 mg/kg连续腹腔注射7 d建立帕金森病(PD)模型。造模完成后MPTP + 天麻素组给予天麻素90 mg/kg灌胃,MPTP组灌胃等体积蒸馏水。连续给药9周,于第8周进行旷场实验。旷场实验记录总路程、平均速度、静止时间、中心区停留时间、中心区进入次数及潜伏期等指标,并结合活动轨迹图进行运动模式分析。结果:MPTP组轨迹减少,主要沿边活动;MPTP + 天麻素组活动范围扩大,中心区进入增加。两组旷场行为指标比较中,总路程、平均速度、静止时间、中心区进入次数及中心区停留时间差异均无统计学意义(P > 0.05)。结论:基于运动轨迹与行为学评价,天麻素能够在一定程度上改善MPTP模型小鼠的探索行为与运动表现,但量化指标差异未达到统计学显著性。
Abstract: Objective: Exploring the protective effects of gastrodin on motor function in mice with parkinson’s disease induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). Method: SPF-grade male C57BL/6 mice were randomly assigned to either the MPTP PD model group or the MPTP + gastrodin group, with 5 mice in each group. A Parkinson’s disease (PD) model was established by continuous intraperitoneal injection of MPTP at 25 mg/kg for 7 days. After model establishment, the MPTP + gastrodin group received gastrodin at 90 mg/kg via oral administration, while the MPTP group received an equal volume of distilled water orally. The drug was administered continuously for 9 weeks, and the open-field test was conducted in the 8th week. The open-field test recorded indicators such as total distance traveled, average speed, stationary time, central zone dwell time, number of central zone entries, and latency period. Additionally, movement pattern analysis was performed using activity trace plots. Results: The MPTP group exhibited reduced movement trajectories, predominantly confined to periphery; the MPTP + gastrodin group demonstrated expanded activity range with increased central zone entry. Comparative analysis of open-field behavioral parameters revealed no statistically significant differences between the two groups in total distance traveled, average speed, rest duration, central zone entry frequency, or central zone dwell time (P > 0.05). Conclusion: Based on movement trajectory and behavioral assessments, gastrodin partially improved exploratory behavior and motor performance in MPTP model mice, though the quantitative differences were not statistically significant.
