HIF-1α在慢性气道炎症性疾病中作用机制的研究进展
Research Progress on the Mechanism of HIF-1α in Chronic Airway Inflammatory Diseases
摘要: 慢性气道炎症性疾病以持续性炎症、气流受限、黏液潴留及气道结构改变为主要特征,其中气道局部低氧微环境可与炎症反应、免疫代谢和组织重塑相互作用,共同促进疾病进展。缺氧诱导因子-1α (HIF-1α)作为低氧应答的核心转录因子,不仅调控细胞对缺氧的适应,也通过炎症因子、糖酵解代谢、免疫细胞活化和血管生成等过程参与慢性气道炎症持续化。结合相关文献,本综述对HIF-1α在支气管哮喘、慢性阻塞性肺疾病(COPD)及其他慢性气道炎症性疾病中的研究进展进行梳理,并讨论其作为候选生物标志物和潜在治疗靶点的价值。
Abstract: Chronic airway inflammatory diseases are mainly characterized by persistent inflammation, airflow limitation, mucus retention and airway structural remodeling. The local hypoxic microenvironment in the airway interacts with inflammatory responses, immune metabolism and tissue remodeling to jointly drive disease progression. As the master transcription factor governing hypoxic response, hypoxia-inducible factor-1α (HIF-1α) not only mediates cellular adaptation to hypoxia, but also participates in the perpetuation of chronic airway inflammation through multiple processes including inflammatory cytokine secretion, glycolytic metabolism, immune cell activation and angiogenesis. Based on existing literature, this review systematically summarizes research advances of HIF-1α in bronchial asthma, chronic obstructive pulmonary disease (COPD) and other chronic airway inflammatory disorders, and discusses its potential value as a candidate biomarker and therapeutic target.
