Toxicity Management in CAR-T Cell Therapy

摘要: 所称的嵌合抗原受体T淋巴细胞(CAR-T)疗法是血液肿瘤治疗中的一项重大进展，但目前因细胞因子释放综合征(CRS)或免疫效应细胞有关的神经毒性(ICANS)而受到一定限制。文中对CAR-T治疗引发的各种毒性作较完整的分析，即对其发生原因、临床特征、分级方法与整体处理做归纳，亦将临床安全纳入考虑范围。

Abstract: Chimeric antigen receptor T-cell (CAR-T) therapy represents a significant advancement in the treatment of hematological malignancies. However, its widespread clinical application remains constrained by treatment-related toxicities, notably cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS). This review provides a comprehensive analysis of the various toxicities associated with CAR-T therapy, encompassing their pathogenesis, clinical manifestations, grading systems, and integrated management strategies, with an emphasis on ensuring clinical safety.
