摘要: 阿尔茨海默病(Alzheimer’s Disease, AD)是一种病因复杂的神经退行性疾病,近年来流行病学与分子生物学研究提示,病毒感染可能在其发病与进展中发挥关键作用。本文从单纯疱疹病毒1型、巨细胞病毒、EB病毒及新型冠状病毒等多种病毒与AD关联的流行病学证据来进行综述,并深入探讨了病毒诱导的神经炎症、β-淀粉样蛋白(β-Amyloid protein, Aβ)沉积、Tau蛋白过度磷酸化及突触功能障碍等核心病理机制。通过整合现有研究,本文旨在阐明病毒感染的致病作用,并评估抗病毒治疗与疫苗接种在AD预防中的潜在价值,从而为AD的病因学探索与临床干预提供新视角。
Abstract: Alzheimer’s Disease (AD) is a neurodegenerative disorder with complex etiology. Recent epide- miological and molecular biological studies suggest that viral infections may play a pivotal role in its pathogenesis and progression. This review synthesizes epidemiological evidence linking various viruses—including herpes simplex virus type 1, cytomegalovirus, Epstein-Barr virus, and SARS-CoV-2—to AD, and delves into core pathological mechanisms such as virus-induced neuroinflammation, β-Amyloid protein (Aβ) deposition, excessive phosphorylation of Tau protein, and synaptic dysfunction. By integrating existing research, this study aims to elucidate the pathogenic role of viral infections and evaluate the potential value of antiviral therapy and vaccination in AD prevention, thereby providing new insights for etiological investigation and clinical intervention strategies of AD.
