Abstract: Pillararenes are a class of macrocyclic host molecules featuring rigid pillar-shaped structures, electron-rich cavities, and readily functionalized rims. Owing to these structural characteristics, pillararenes have attracted considerable attention in molecular recognition, supramolecular assembly, and fluorescence regulation. This review summarizes recent advances in pillararene-based fluorescent systems, focusing on major construction strategies, including covalent modification with fluorophores, host-guest assembly, aggregation-induced emission, artificial light harvesting, and solid-state materials. The effects of spatial preorganization, host-guest recognition, and local microenvironment regulation on the fluorescence properties of these systems are discussed, together with the major fluorescence response mechanisms, including photoinduced electron transfer (PET), fluorescence resonance energy transfer (FRET), aggregation-induced emission (AIE), and aggregation-caused quenching (ACQ). Their applications in ion and small-molecule sensing, sensor arrays, bioimaging, drug delivery, luminescent materials, and photocatalysis are also summarized. Finally, the current development of pillararene-based fluorescent systems is summarized, and their prospects in sensing, biomedical applications, and photofunctional materials are discussed.
