Abstract: The tumor microenvironment (TME) is a complex ecosystem in which tumor cells survive and develop, and abnormalities in its composition and function serve as important drivers of tumor initiation, progression, metastasis, and drug resistance. With the rapid development of technologies such as single-cell sequencing and spatial transcriptomics, understanding of the TME has evolved from the traditional concept of a cellular community to a complex system encompassing multiple dimensions, including cellular components, the extracellular matrix, vascular networks, and metabolic features. This article systematically reviews the multidimensional characteristics of the TME, summarizes progress on targeted drugs directed against features of each dimension, and introduces emerging frontiers such as cell therapy, the tumor microbiome, and interventions targeting physical barriers. Furthermore, it discusses the challenges and future directions in this field, with the aim of providing a reference for the development of TME-targeted drugs.
