灵芝多糖提取工艺研究

摘要: 本文主要研究了两种灵芝多糖的最佳提取工艺,比较两种灵芝多糖含量的不同。采用热水浸提法对灵芝多糖的提取工艺条件进行研究,以多糖得率为评价指标,选用单因素试验和L18(35)正交试验法,考察浸提比、pH值、浸提温度、乙醇浓度、浸提时间等5个因素对灵芝多糖得率的影响。确定了热水浸提法提取灵芝多糖的最佳工艺条件是浸提比1:40、pH 7.5、浸提温度90℃、浸提时间1.5 h、乙醇浓度90%。采用Sevage法除游离蛋白,3.5-二硝基水杨酸(DNS)法测定灵芝多糖含量。
Abstract: In this paper, the optimum extraction process of two kinds of Ganoderma lucidum polysaccharide was studied, and the content of two kinds of Ganoderma lucidum polysaccharide was compared. The hot water extraction method is used to study the conditions of extracting polysaccharides from Ganoderma lucidum. With taking the polysaccharide yield as the evaluation index, the single factor test and L18 (35) orthogonal test are used to examine the effect of the extraction ratio, pH value, extraction temperature, ethanol concentration and extraction time on the yield of Ganoderma lucidum polysaccharides. The optimum processing conditions for Ganoderma lucidum polysaccharides extraction with hot water are determined as that of extraction ratio 1:40, pH 7.5, extraction temperature 90˚C, extraction time 1.5 h, and ethanol concentration 90%. The content of Ganoderma lucidum polysaccharide was determined by Sevage method with the removal of free protein and 3,5-Dinitrosalicylic acid (DNS).
