地胆草中羽扇豆醇的提取及采用反相HPLC法测定其含量
The Extraction and Quantitative Analysis of Lupeol of Elephantopus scaber Linn. by RP-HPLC
摘要:
我们采用化学提取法从地胆草中提取了羽扇豆醇,并建立了反相-HPLC法测定地胆草中羽扇豆醇含量的方法,为地胆草的质量分析和评价提供科学依据。采用了Wondasil C18柱(4.6 mm × 150 mm, 5 µm),柱温为30˚C,甲醇-水(9:1)为流动相,在流速1.0 mL∙min−1和紫外检测波长206 nm下进样20 µL对地胆草石油醚提取物进行HPLC分析。结果发现:羽扇豆醇在0.25~5.0 µg∙mL−1 (r = 0.9998)呈良好的线性关系,平均加样回收率为98.1% (RSD 1.87%)。本方法简便,重复性好,为评价地胆草药材的质量提供科学依据。
Abstract:
The lupeol was extracted from Elephantopus scaber Linn. by the chemical extraction process and the content of lupeol in Elephantopus scaber Linn. was detected by the RP-HPLC method for the quantitative analysis of lupeol of Elephantopus scaber Linn. The chromatographic parathion was accomplished on a Wondasil C18 column (4.6 mm × 150 mm, 5 μm) eluted with a mobile phase composed of methanol and water (9:1), and with a flow rate of 1.0 mL•min−1. The RP-HPLC expe-riment was achieved at 30˚C, and UV detection was set at 206 nm. The calibration curve was linear in the range of 0.25~5.0 µg∙mL−1 (r = 0.9998) for lupeol, with acceptable precision and accuracy. The established method was proved to be simple, reproducible, accurate, and can be used to evaluate the quality of Elephantopus scaber Linn.
