FAK/PI3K/AKt/mTOR信号通路相关蛋白在胃癌中的表达及意义
The Expression and Significance of FAK/PI3K/AKt/mTOR Signaling Pathway Related Proteins in Human Gastric Carcinoma

目的:探讨FAK、PI3K、AKT、mTOR蛋白在胃癌组织中表达与临床病理及生存的关系。方法:应用免疫组化的方法检测50例胃癌组织、48例癌旁组织、20例正常胃组织中FAK、PI3K、p-AKT、p-mTOR蛋白的表达。结果:FAK、p-AKt、p-mTOR在胃癌组织中的表达高于癌旁组织、正常胃组织(P < 0.01),其表达水平与浸润深度、临床分期、淋巴结转移呈正相关(P < 0.01, r = 0.81),与组织分化类型呈负相关(P < 0.01, r = −0.85),与患者的性别、年龄、胃癌的部位无相关(P > 0.05);PI3K在胃癌组织中的表达高于癌旁组织、正常胃组织(P > 0.05),其表达水平与患者的性别、年龄、胃癌的部位、浸润深度、临床分期、淋巴结转移无相关(P > 0.05);生存分析显示,p-AKt、p-mTOR阳性表达与阴性表达患者的生存时间比较差异有统计意义(P < 0.01);FAK、PI3K阳性表达与阴性表达患者的生存时间比较差异无统计意义(P > 0.05)。结论:AKT、mTOR的异常表达参与了胃癌的演进过程,可能是胃癌患者预后的一个独立指标之一。
Abstract:
Object: To explore the relationship between the expression of FAK, PI3K, AKt and mTOR protein in gastric carcinoma and analyze the relationship with clinicopathology and survival. Methods: Im-munohistochemical method was used to detect the level of FAK, PI3K, p-AKt and p-mTOR protein in 50 cases of gastric carcinoma, 48 cases of tissue adjacent to carcinoma and 20 normal gastric tissues. Result: The level of FAK, p-AKt and p-mTOR in gastric cancer carcinoma was higher than that of tissue adjacent to carcinoma and normal gastric tissues (p < 0.01), its level was positively correlated with infiltration depth, clinical stage, lymph node metastasis (P < 0.01), and negatively correlated with the type of tissue differentiation (P < 0.01), was not correlated with the gender, age and gastric carcinoma area of patients ((P > 0.05); the level of PI3K in gastric carcinoma was higher than that of tissue adjacent to carcinoma and normal gastric tissue (P > 0.05), and the level was not correlated with the patient's gender, gastric carcinoma area, infiltration depth, clinical stage and lymph node metastasis (P > 0.05). Survival analysis shows, the survival time of patients with p-AKt, p-mTOR positive expression and negative expression was statistically significant (p < 0.01), and with FAK, PI3K negative expression was not statistically significant. Conclusion: The abnormal expression of AKt and mTOR is involved in the evolution of gastric carcinoma, which may be one of the independent indexes of prognosis of gastric carcinoma patients.
