黄瓜香对高脂饲料诱导的大鼠非酒精性脂肪肝保护作用研究

摘要: 目的:研究高脂饲料诱导大鼠非酒精性脂肪肝模型及黄瓜香提取物的保护作用(nonalcoholic fatty liver disease, NAFLD),并初步探讨其相关作用机制。方法:健康雄性大鼠45只,随机分为3组:对照组、模型组和黄瓜香提取物组(7.5 g∙kg−1∙d−1),除对照组用普通饲料喂养外,其余各组均给予高脂饲料喂养。实验10周后处死全部大鼠,比较各组之间血清和肝脏生化以及肝脏组织病理学特征的差异。结果:与对照组相比,模型组大鼠血清甘油三酯(TG)、胆固醇(TC)、血清丙氨酸转氨酶(ALT)、门冬氨酸转氨酶(AST)含量和肝组织丙二醛(MDA)含量显著升高(P < 0.01),肝组织超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)含量显著降低(P < 0.01);黄瓜香提取物组大鼠血清TC、TG、ALT、AST和肝组织MDA含量显著低于模型组(P < 0.05),肝组织SOD和GSH-Px的含量显著高于对照组(P < 0.01);模型组大鼠肝脏脂肪变性严重,并伴有炎细胞浸润及坏死;黄瓜香提取物组大鼠肝脏脂肪变性程度轻,无明显炎细胞浸润及坏死。结论:黄瓜香提取物对高脂饲料诱导的大鼠非酒精性脂肪肝有明显的保护作用。
Abstract: AIM: To investigate the protective effect of Viola diffusa Ging (VDG) on nonalcoholic fatty liver disease in rats. METHODS: 45 adult male rats, weighing 180-200 grams, were randomly divided into three groups after adaptability keeping five days. The normal group fed with basic animal feeds; fatty liver model group fed with high fat diet. VDG group fed with high fat diet + Kiwi Essence; the body weight, liver index, the serum TG, TC, ALT, AST and hepatic TG, TC, MDA, SOD, GSH-PX were detected, and the histological changes in liver (H.E. staining and oil red staining) were evaluated. RESULTS: Compared with the control group, the liver index (3.73% ± 0.19%), the serum TG (4.97 ± 0.55 mmol/L), TC (6.65 ± 0.23 mmol/L), ALT (79.36 ± 22.61 U/L), AST (177.36 ± 22.15), hepatic MDA (13.48 ± 2.32 U/mg pro) of the model group were markedly higher (P < 0.01), the hepatic SOD (96.82 ± 20.09 U/mg pro) and the hepatic GSH-PX (42.53 ± 9.97 U/mg pro) were significantly lower (P < 0.01). Compared with the model group, the liver index, the serum TG, TC ALT, AST, hepatic MDA of the Kiwi Essence middle-dose and high-dose group were markedly lower (P < 0.01); the hepatic SOD and the hepatic GSH-PX were significantly higher (P < 0.01). Liver pathology showed: Normal group were not unusual; liver steatosis of fatty liver model group was more serious and with infiltration of inflammatory cells and necrosis; liver steatosis of the VDG group was lighter. CONCLUSION: VDG has a protective effect on fatty liver of rat.
