Detection of Virulence Factor Delta by PCR and Biochemical Methods to Identify Heat-Labile Enterotoxigenic Escherichia coli-Caused Diarrhoea
摘要: 目的:探讨PCR及生物化学方法对确定ETEC的分子特性和毒力谱的重要意义,并通过检测毒力基因eltA的存在诊断热不稳定(LT)肠毒素产生的ETEC导致的腹泻。方法:采用RT-PCR方法,确认两株大肠杆菌为热不稳定的ETEC。被动胶乳凝集实验将被用来测试样品中LT毒素的存在。结果:在这项研究中,样本1和样本3均表达eltA基因,乳胶凝集显示样本3的呈阳性。结论:染色结果提示此腹泻儿童极有可能感染了热不稳定的ETEC。
Abstract: Objective: To investigate expressions and clinical significance of determining the molecular characteristic and identify the virulence profile of the ETEC and heat-labile (LT) enterotoxin producing ETEC can be identified by examining the presence of the virulence gene eltA. Methods: By using RT-PCR, two E. coli strains were confirmed to be the LT producing ETEC. The passive latex agglutination was allowed to test the presence of the LT toxin in the specimens. Results: In this study, both samples 1 and 3 specifically expressed the eltA gene, the sample 3 from child with diarrhoea was positive for the latex agglutination. Conclusion: It was highly likely that child was infected by LT producing ETEC stain.
