HPLC法测定板蓝根颗粒提取物的尿苷、鸟苷及腺苷含量

摘要: 目的:采用高效液相色谱法(HPLC)建立板蓝根颗粒提取物含量测定的质量控制方法。方法:采用C18 4.6 × 250 mm色谱柱(天津喆分医药科技、中谱科技和谱宁科技),甲醇和水的梯度洗脱,流速0.8 ml/min,检测波长为254 nm,柱温为30℃。结果:尿苷在5.05 μg/ml~80.88 μg/ml范围内有良好的线性关系;鸟苷在5.01 μg/ml~80.08 μg/ml范围内有良好的线性关系;腺苷在6.23 μg/ml~99.60 μg/ml范围内有良好的线性关系,尿苷回归方程y = 0.8851x − 0.1210 (r2 = 1.000),鸟苷回归方程y = 1.0852x − 0.1685 (r2 = 1.000);腺苷回归方程y = 1.6328x – 02553 (r2 = 1.000)。尿苷回收率为102.6%,RSD为1.2%;鸟苷回收率为99.1%,RSD为0.9%;腺苷回收率为98.8%,RSD为1.0%,回收率良好。结论:该方法科学、简便、准确,能有效控制板蓝根颗粒提取物含量测定的质量。
Abstract: Objective: To establish a high performance liquid chromatography (HPLC) for the quality control of Radix Isatidis granule extract. Methods: A C18 4.6 × 250 mm chromatographic column (Tianjin Zhefen Medicine Science and Technology Co., Ltd., Zhongpu Technology Co., Ltd. and Puning Technology Co., Ltd.), and gradient elution of methanol and water were used, the flow rate was 0.8 ml/min, the detection wavelength was 254 nm, and the column temperature was 30˚C. Results: Uridine had a good linear relationship in the range of 5.05 μg/ml~80.88 μg/ml. Guanosine has a good linear relationship in the range of 5.01 μg/ml~80.08 μg/ml. Adenosine ranged from 6.23 μg/ml to 99.60 μg/ml, uridine regression equation y = 0.8851x − 0.1210 (r2 = 1.000), guanosine regression equation y = 1.0852x − 0.1685 (r2 = 1.000); The regression equation of adenosine is y = 1.6328x − 02553 (r2 = 1.000). The recovery rate of uridine was 102.6%, RSD was 1.2%. The recovery rate of guanosine was 99.1% and RSD was 0.9%. The recovery rate of adenosine was 98.8%, RSD was 1.0%, and the recovery rate was good. Conclusion: The method is scientific, simple and accurate, and can effectively control the quality of the content determination of Radix Isatidis granule extract.
