摘要: DNA损伤修复(DNA damage repair, DDR)系统在保护基因组稳定性方面发挥重要作用,DNA损伤累积或DDR功能减弱都会导致肿瘤的发生发展,同时也给肿瘤的药物治疗提供机会和靶点。人类X射线修复交叉互补基因1 (X-ray repair cross complementing gene 1, XRCC1)是首个发现影响细胞对电离辐射敏感性的哺乳动物类基因,调控DNA单链断裂和碱基切除损伤修复通路中的关键步骤。本文就XRCC1的结构、功能及在恶性肿瘤中的最新研究成果作一简要综述。
Abstract: DNA damage repair (DDR) system plays an important role in maintaining genomic stability. The accumulation of DNA damage or the weakening of DDR function will lead to the occurrence and de-velopment of tumors, and also provide opportunities and targets for drug therapy of tumors. Hu-man X-ray repair cross complementing gene 1 (XRCC1) is the first mammalian gene found to affect cell sensitivity to ionizing radiation, and plays an important role in regulating DNA single-strand break and base excision damage repair pathways. This article reviews the structure and function of XRCC1 and the latest research results in malignant tumors.
