摘要: 目的:探讨三四脯氨酸(Tristetraprolin, TTP)对小鼠烟曲霉菌性角膜炎中炎症反应的影响。方法:体外实验用烟曲霉菌菌丝感染RAW264.7细胞后,用RT-PCR方法检测RAW264.7细胞中不同时间段TTP、IL-1β、TNF-α、IL-6和CCL-2的mRNA表达。使用靶向TTP表达的短干扰RNA (TTP siRNA)或乱序干扰RNA (scrambled RNA)处理,干扰TTP的表达。RT-PCR检测干扰TTP表达后烟曲霉菌诱导的RAW264.7中IL-1β、TNF-α、IL-6,CCL-2的mRNA的表达。结果:TTP在烟曲霉菌刺激的RAW264.7细胞中表达增加,TTP siRNA可以抑制RAW264.7细胞中TTPmRNA的表达,干扰TTP的表达增加了烟曲霉菌诱导的RAW264.7细胞炎症反应中的IL-1β、IL-6和CCL-2的表达。结论:TTP抑制烟曲霉菌诱导的RAW264.7细胞炎症因子表达。
Abstract: Objective: To explore the effect of Tristetraprolin (TTP) on Aspergillus fumigates (A. fumigatus) ker-atitis in mice. Methods: After RAW264.7 cells were infected with Aspergillus fumigatus mycelium in vitro, the expression of TTP mRNA in RAW264.7 cells was detected by RT-PCR. Then, RAW264.7 cells were treated with short interfering RNA (TTP siRNA) targeting TTP expression or out-of-order interfering RNA (scrambled RNA) and the mRNA expression of TTP, IL-1β, TNF-α, IL-6 and CCL-2 by RT-PCR. Results: The expression of TTP increased in RAW264.7 cells stimulated by Aspergillus fu-migatus. TTP siRNA could inhibit the expression of TTP mRNA in RAW264.7 cells. Interfering the expression of TTP increased the expression of IL-1β, IL-6 and CCL-2 in the inflammatory reaction of RAW264.7 cells induced by Aspergillus fumigatus. Conclusion: TTP can inhibit the expression of in-flammatory factors in RAW264.7 cells induced by Aspergillus fumigatus.
