低氧条件下HIF对间充质干细胞成骨分化影响的研究

摘要: 骨不连和骨缺损是创伤骨科的重要问题。通过干细胞成骨分化来修复骨缺损及骨不连,将彻底改变传统的通过创伤较大的手术方式修复骨缺损的现状,迈入无创伤修复的新阶段。研究表明,氧环境切换可能会影响干细胞分化潜能,本文综述了近年来关于缺氧诱导因子(HIF)对于缺氧条件下的细胞调控,得出了HIF-2α可能是MSCs成骨分化的新解决方案这一结论。HIF-2α作为与慢性低氧环境调节相关的核心转录因子,在间充质干细胞增殖和分化中发挥重要作用。
Abstract: Bone nonunion and bone defect are important problems in traumatic orthopedics. Osteogenic dif-ferentiation of stem cells to repair bone defects and bone nonunion will completely change the tra-ditional situation of repairing bone defects through more traumatic surgery, and enter a new stage of non-traumatic repair. Studies have shown that oxygen environment switching may affect the dif-ferentiation potential of stem cells. In this paper, we reviewed the recent studies on hypox-ia-inducing factor (HIF) regulation of cells under hypoxia conditions, and concluded that HIF-2α may be a new solution for osteogenic differentiation of MSCs. HIF-2α, as a core transcription factor relat-ed to the regulation of chronic hypoxic environment, plays an important role in the proliferation and differentiation of mesenchymal stem cells.
